EZ DNA Methylation-Lightning

The EZ DNA Methylation-Lightning Kits은 빠르고 안정적인 bisulfite conversion을 지원하는 methylation analysis용 제품으로, 약 1.5시간 이내에 DNA의 bisulfite conversion 및 purification이 가능합니다. 본 제품의 가장 큰 특징은 별도의 시약 준비 과정 없이 바로 사용할 수 있는 Ready-to-use Lightning Conversion Reagent를 제공한다는 점으로, DNA sample에 직접 첨가 후 약 1시간 정도 incubation만 진행하면 conversion이 완료됩니다. 또한 Converted DNA의 desulphonation 및 clean-up 과정은 low-elution spin column 기반으로 수행되며, 최소 10 µl 수준의 low-volume elution이 가능합니다. 높은 cytosine to uracil conversion efficiency를 제공하여 이후 PCR, Array, Bisulfite Sequencing 및 NGS와 같은 downstream methylation analysis에 적합합니다.

PCR-Ready Bisulfite DNA from FFPE

DNA isolated from a non-small-cell lung cancer (NSCLC) FFPE block was bisulfite converted using the EZ DNA Methylation-Lightning Kit, then PCR amplified with methylation-specific primers for cancer biomarkers SEPT9, RASSF1A, Mir129-2, and control primer COL2A1. L=50bp marker.

Recovery of Small Fragments

Bisulfite converted libraries of small 100, 200, and 300bp DNA fragments were successfully recovered and amplified by PCR. Libraries were analyzed using the Agilent 4200 TapeStation system. 

High recovery rates of bisulfite converted fragmented DNA were observed through qPCR analysis targeting the C-LESS-C1 region (∆ct=0.945). Fragment size prior to bisulfite conversion averaged 175bp.
